Protein/peptide accurate mass determination by ESI or MALDI

What you need to provide:

  1. Salt/detergent free sample at a concentration of at least 5 pmole/ml in a volatile solvent (water or acetonitrile). Lyophilized samples are preferable.
  2. Include information about exact sequence of peptide/protein, solubility properties, and expected MW, whenever possible.
  3. Duly completed sample submission form and Waiver/disclaimer forms.

Protein Identification by LC-tandem MS or MALDI

What you need to provide:

For protein identification by either LC-tandem MS or MALDI, PCL will carry out the in-gel digestion, extraction of peptides, data acquisition, interpretation of the data. Generally, if you can see the protein by Coomassie, we can identify it. Hence, Coomassie stained gel spot/bands are preferred. If you have to do silver or sypro ruby staining, please follow the instructions mentioned in the General Protocols. Excise the gel plug using a clean scalpel and put it in a 0.5 ml Eppendorf tube (pre-cleaned with methanol or acetonitrile). No buffer/destainer needed. Please ship all materials on dry ice by overnight carrier.

Required information with submitted sample:

  1. We prefer that the sample be accompanied by a picture of the whole gel with the band/spot of interest clearly marked.
  2. The sample submission form with all the relevant information - source of the sample, status of cysteines, pI and MW (if known) etc. - clearly printed.
  3. Waiver/disclaimer form.
    Note: These forms can be submitted just once per year.
